Product Specific Information

The monoclonal antibody IgE21 recognizes the immunoglobulin E (IgE) class of antibodies. IgE levels in serum are typically low and play a crucial role in allergic responses by binding to mast cells and basophils via Fc epsilon receptor I, leading to histamine release. IgE is produced by plasma B cells and basophils and exists in both soluble and membrane-bound forms. IgE21 recognizes both forms.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and validated for flow cytometric analysis of normal human peripheral blood cells.
Use 5 µL (0.2 µg) per test in a final volume of 100 µL. The cell number per test can range from 10⁵ to 10⁸ cells.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes such as propidium iodide, 7-AAD, or DAPI.
• Use Invitrogen LIVE/DEAD Fixable Dead Cell Stains for compatibility.
• This conjugate ships with CellBlox Plus Blocking Buffer (Cat. No. C001T06F01), which offers improved formulation over the original CellBlox.
• Add 5 µL of CellBlox Plus Blocking Buffer per sample before adding cells.
• For single-color controls, use 5 µL per 100 µL of cell sample containing 10³–10⁸ cells.

Target Information

IgE is one of five immunoglobulin classes in mammals, composed of two epsilon heavy chains and two light chains. It is essential in type I hypersensitivity reactions (e.g., hay fever, asthma, hives, anaphylaxis) by binding Fc receptors on basophils and mast cells. Crosslinking of receptor-bound IgE triggers degranulation and release of allergic mediators. IgE also contributes to parasite immunity.
