Target Information
Peroxiredoxin (Prx) is a growing peroxidase family whose mammalian members are associated with cell proliferation, differentiation, and apoptosis. Approximately 50 isoforms exist, classified by amino acid sequence homology and active site cysteine residues, exhibiting thiol-specific antioxidant activity across all kingdoms.
Mammalian Prx includes six members grouped into typical 2-Cys, atypical 2-Cys, and 1-Cys Prx types. Except for Prx VI (1-Cys subgroup), the other five 2-Cys Prx isotypes possess thioredoxin-dependent peroxidase (TPx) activity utilizing thioredoxin, thioredoxin reductase, and NADPH as a reducing system.
Mammalian Prxs are 20–30 kDa in molecular size and vary in subcellular localization:
- Prx I, II, VI: Cytosol
- Prx III: Mitochondria
- Prx IV: Endoplasmic Reticulum and secretion
- Prx V: Peroxisome, mitochondria, and cytosol
