The eBioR2/60 monoclonal antibody reacts with mouse CD43 (leukosialin, sialophorin). CD43 glycoforms (~115 and 130 kDa) are expressed by early B cell subsets in bone marrow, thymocytes, mature T cells, NK cells, myeloid cells, and platelets. CD43 binds to CD54 and plays roles in adhesion and B cell survival.
Clone R2/60 detects both isoforms of CD43, unlike S7 and 1B11 clones that recognize distinct isoforms. R2/60 stains most CD4+, CD8+, and CD19+ splenocytes, indicating recognition of a shared epitope across CD43 isoforms.
Applications Reported: Flow cytometry, immunoprecipitation, and immunoblotting (WB).

Applications Tested: Flow cytometric analysis of mouse splenocytes at ≤0.5 µg/test (100 µL final volume). Optimal performance requires empirical titration.

Target Information
CD43 (leukosialin, sialophorin) is a transmembrane mucin-like protein expressed on most hematopoietic cells. It contributes to a repulsive barrier affecting cellular adhesion and can promote leukocyte aggregation. Through interaction with actin-binding proteins (ezrin, moesin), CD43 regulates T-cell morphology and lymphocyte traffic.
CD43 signaling modulates LFA-1 adhesiveness and influences tumor-related pathways involving p53. Dysfunction of CD43 is associated with Wiscott-Aldrich Syndrome and Adenoid Basal Cell Carcinoma.
