Description:
The HIP1 monoclonal antibody reacts with the human CD42b molecule, a 135 kDa transmembrane protein also known as platelet gpIb.
Applications Reported:
HIP1 antibody has been reported for use in:
- Flow cytometric analysis
- In vitro blocking of human CD42b
- Immunoblotting (WB)
- Immunohistochemical staining
(For functional assays, use Functional Grade purified HIP1, Product #16-0429.)
Applications Tested:
Tested by flow cytometric analysis of human platelets. Recommended ≤1 µg per test in 100 µL final volume. Cell number may range from 10⁵ to 10⁸ cells/test. Optimize antibody titration for best performance.
Purity: Greater than 90%, determined by SDS-PAGE.
Aggregation: Less than 10%, determined by HPLC.
Filtration: 0.2 µm post-manufacturing filtered.
Target Information
CD42b is a platelet activation marker involved in coagulation and platelet aggregation. It interacts with extracellular matrix and adhesion molecules, and is regulated by VEGF and PDGF. Together with GPIb beta, GPIX, and GPV, CD42b forms the GPIb-IX-V receptor complex, which mediates platelet tethering to thrombogenic surfaces and binds von Willebrand factor (VWF) at vascular injury sites. It also interacts with thrombin, coagulation factors XI and XII, high molecular weight kininogen, TSP-1, integrin Mac-1, and P-selectin. CD42b expression occurs on platelets and bone marrow megakaryocytes. Mutations in CD42b are associated with Bernard-Soulier syndrome and platelet-type von Willebrand disease.
