Product Specific Information

The monoclonal antibody R26.4C recognizes the tight junction protein ZO-1 (zona occludens) in rat, mouse, canine, and porcine.
ZO-1 is a member of the MAGUK family of tight junction–associated scaffolding proteins, containing PDZ, SH3, and GUK domains. It is an integral component of the tight junction complex, forming a barrier to paracellular movement of substances and maintaining epithelial cell polarity.

ZO-1 interacts with ZO-2, ZO-3, occludin, cingulin, claudins, actin, alpha-catenin, AF-6, ZONAB, Apg-2, and JAM-A–C, anchoring the actin cytoskeleton to the tight junction.
Down-regulation of ZO-1 and ZO-2 has been associated with cancer progression.

Applications Reported:
This antibody is suitable for immunohistochemical staining of frozen tissue sections, microscopy, and immunocytochemistry.

Applications Tested:
Validated for immunocytochemistry of formaldehyde-fixed and permeabilized cells at ≤1 µg/mL. Optimal dilution should be determined experimentally.

Filter Recommendation:
Use a filter capturing the 570 nm emission wavelength (e.g., Excitation 545/25, 565LP, Emission 605/70).
Standard Alexa Fluor® 555 or TRITC filters are acceptable.

Target Information

ZO-1 forms complexes with ZO-2 or ZO-3 and associates with claudin, occludin, and F-actin at tight junction strands, linking the actin cytoskeleton to the junction.
ZO-1 expression is reduced in many breast cancer lines.
ZO-2 and ZO-3 are ubiquitously expressed in epithelial tight junctions, while ZO-1 is also expressed at cardiac myocyte junctions.
ZO-1 may be involved in signal transduction at cell-cell junctions.
Two transcript variants encoding distinct isoforms have been identified.
Diseases associated with ZO-1 dysfunction include Celiac Disease and Congenital Nephrotic Syndrome.
