Product Specific Information

This ICACLS monoclonal antibody reacts with human, mouse, and rat glucagon. Glucagon is a peptide hormone secreted by pancreatic alpha cells within the islets of Langerhans. It functions as an antagonist to insulin, maintaining blood glucose homeostasis through glycogenolysis and gluconeogenesis in the liver.
Secretion is stimulated by hypoglycemia, epinephrine, acetylcholine, and cholecystokinin, and inhibited by insulin and somatostatin. Elevated glucagon levels are often found in pancreatic tumors.

Applications Reported:
Used for immunohistochemical staining of formalin-fixed paraffin-embedded tissue sections.

Applications Tested:
Tested by IHC using low pH antigen retrieval at ≤20 µg/mL. Antibody should be titrated for optimal performance.

Filter Recommendation:
Use filters capturing 570 nm emission (e.g., Excitation 545/25, 565LP, Emission 605/70). Standard Alexa Fluor® 555 or TRITC filters are acceptable.

Target Information

Glucagon is a 29-residue polypeptide hormone (MW 3482) produced in the pancreas. Enteroglucagon (oxyntomodulin), produced in intestinal mucosa, extends glucagon’s sequence by 8 residues.
Biological activities include glycogenolysis, lipolysis, gluconeogenesis, and ketogenesis—antagonistic to insulin action, increasing blood glucose levels.
Immunocytochemical studies show glucagon localized in alpha cells (15–20% of islet cells), primarily at the islet periphery.
Glucagon-specific antibodies are useful as cell/tumor markers and analytical tools for hormone quantification.
