Product Specific Information

This 4SM95 monoclonal antibody reacts with the mouse CD4 molecule, a 55 kDa cell surface receptor expressed by thymocytes, mature T cells, and dendritic cells. CD4 binds to MHC class II on antigen-presenting cells and is essential for T cell development and function. In T cells, CD4 associates with protein tyrosine kinase p56lck through its cytoplasmic tail.

The 4SM95 antibody is not recommended for immunohistochemistry of frozen mouse tissue. For frozen tissue applications, refer to alternate products:

Applications Reported:
Immunohistochemical staining of formalin-fixed paraffin-embedded tissue sections and microscopy.

Applications Tested:
Tested by IHC on formalin-fixed paraffin-embedded mouse tissue using low pH antigen retrieval. Recommended use ≤ 20 µg/mL. Optimize dilution for best results.

Filter Recommendation:
For eFluor® 570 conjugate, use filters capturing 570 nm emission (e.g., Excitation 545/25, 565LP, Emission 605/70). Alexa Fluor® 555 or TRITC filters are acceptable.

Target Information

CD4 antigen recognizes MHC class II molecules and acts as a co-receptor for HIV. Primarily expressed in T helper cells, but also found in monocytes, macrophages, and dendritic cells. Tissue expression includes thymus, lymph nodes, tonsils, spleen, and select regions of brain and gut. CD4 facilitates T-cell activation via association with TCR complex and Lck kinase, and may mediate neuronal damage in immune-related CNS diseases. Multiple alternatively spliced transcripts identified (RefSeq, July 2017).
