Product Specific Information

The 6C2 monoclonal antibody reacts with mouse Toll-like receptor 2 (TLR2). Mouse TLR2 is expressed by the myeloid lineage, including macrophages and dendritic cells in splenocytes and the RAW264.7 cell line.
TLR2 and TLR4 are pattern recognition receptors that respond to bacterial lipoproteins and are key in innate immunity and inflammation. TLR2 is a surface receptor responsible for distinguishing different pathogens.

Applications Reported:
6C2 has been reported for use in flow cytometric analysis. For optimal staining, use directly conjugated 6C2 (FITC, PE, or Biotin) with FcR blocking using anti-mouse CD16/CD32 (clone 93, Product #14-0161).

Applications Tested:
Tested by flow cytometric analysis of mouse splenocytes. Use ≤1 µg per test (100 µL final volume). Cell number may range from 10⁵–10⁸ cells/test. Titrate antibody for optimal performance.

Excitation / Emission / Laser:
488–561 nm excitation; 578 nm emission; compatible with Blue, Green, and Yellow-Green lasers.

Filtration:
0.2 µm post-manufacturing filtered.

Target Information

TLR2 is a member of the Toll-like receptor family, crucial for pathogen recognition and activation of innate immunity.
TLR2 forms heterodimers with TLR1 or TLR6 to recognize bacterial and mycoplasma lipoproteins.
Highly conserved across species, TLR2 is abundantly expressed in peripheral blood leukocytes and mediates host responses to Gram-positive bacteria and yeast via NF-kappaB activation.
Expression levels vary by cell type and are modulated by immune response modifiers such as LPS, GM-CSF, IL-1, and IL-10 (upregulation) or IL-4, IFN-γ, and TNF (downregulation).
