Product Specific Information

This SC1T2M3 monoclonal antibody recognizes human and mouse Src tyrosine kinase phosphorylated on tyrosine 418 (Y418). Autophosphorylation at Y418 is essential for full catalytic activity. Src is a non-receptor tyrosine kinase involved in signal transduction across diverse receptor classes including immune receptors, integrins, receptor tyrosine kinases, GPCRs, and cytokine receptors. Aberrant Src activity is implicated in various cancers.

Due to sequence homology around Src Y418, this clone is predicted to cross-react with other Src family kinases (Src, Lck, Fyn, Lyn). Specificity was confirmed by ELISA, flow cytometry, and western blotting.

  • Applications Tested: Pre-titrated for intracellular staining of normal human peripheral blood cells at 5 µL (0.25 µg)/test

Target Information

SRC is a cytoplasmic tyrosine kinase and proto-oncogene regulating embryonic development, cell growth, differentiation, and apoptosis. It associates with cytoplasmic regions of various receptors for signal transduction and participates in cytoskeletal organization and cell cycle control. In humans, SRC is located on chromosome 20.

Phosphorylation of Tyr416 activates SRC, while Tyr527 phosphorylation inhibits it. Mutations in SRC may contribute to malignant progression of colon cancer. Two transcript variants encode the same SRC protein.
