Product Specific Information

This LUVNKLA monoclonal antibody recognizes human and mouse STAT3 phosphorylated at tyrosine 705 (Y705). STAT proteins are transcription factors activated by cytokine receptor-associated JAK kinases, mediating cellular processes such as apoptosis, differentiation, and proliferation.

Following phosphorylation, STAT3 translocates to the nucleus to regulate gene transcription. It is activated downstream of cytokines such as IFNs, IL-5, IL-6, IL-10, and LIF, and is crucial for Th17 differentiation and cell survival. Abnormal STAT3 activity is linked to various cancers and immune dysregulation.

Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Pre-titrated and validated for intracellular staining of human PBMCs. Use 5 µL (0.06 µg) per 100 µL test volume. Cell count: 10⁵–10⁸ cells/test.

Recommended Protocol:
Use “Protocol C: Two-step protocol (Fixation/Methanol)”.
Protocols A and B are not suitable.

Target Information

STAT3 is a member of the STAT family of transcription factors that translocate to the nucleus upon phosphorylation by receptor-associated kinases. It is activated by cytokines and growth factors such as IFN-α, IFN-γ, EGF, PDGF, and IL-6. Phosphorylation at Tyr705 by JAK1/2 is essential for STAT3 dimerization and DNA binding.

STAT3 regulates genes involved in growth, survival, and apoptosis. It interacts with coactivators like CREB-binding protein and is inhibited by PIAS3. The STAT3 gene is located on chromosome 17q, and its deletion is embryonically lethal in mice.
