Specificity Comments:
Actin is a major component of the cytoskeleton and is present in most cell types. This antibody is highly specific to actin from smooth muscle and does not stain cardiac or skeletal muscle. It stains myofibroblasts and myoepithelial cells. Useful in differential diagnosis of smooth and skeletal muscle tumors.

• Rhabdomyosarcoma: negative with this antibody, positive with anti-muscle specific actin and myogenin.
• Leiomyosarcoma: positive with anti-muscle specific actin and anti-smooth muscle actin, negative with anti-myogenin.

Smooth Muscle Actin (α-SMA) is part of the actin protein family involved in cell motility, structure, and integrity.

• Isoforms: α, β, and γ actins; α-actin is a major constituent of the contractile apparatus.
• Localization: Found in smooth muscle vessel walls, gut wall, myometrium, and myoepithelial cells of breast and salivary glands.
• Function: Interacts with myosin for muscle contraction, cell motility, and cytokinesis.
• Clinical Relevance: Mutations in smooth muscle actin gene cause familial thoracic aortic aneurysm type 6.
• Expression: α-SMA is abundant in vascular and visceral smooth muscle cells and appears in fibroblast stress fibers during wound healing and fibrocontractive diseases.
• Variants: Multiple alternatively spliced variants identified.
