Product Specific Information

The G14D9 monoclonal antibody reacts with human Glycoprotein A Repetitions Predominant (GARP, also known as LRRC32 or Garpin). GARP is an approximately 80 kDa glycoprotein expressed on the cell surface. Expression has been reported in placenta, lung, kidney, heart, ovary, liver, skeletal muscle, and pancreas. Protein expression is observed on megakaryocytes, platelets, and activated regulatory T (Treg) cells. GARP expression on activated Treg cells is necessary for their suppressive function and may act as a receptor for LAP/TGF beta.

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-titrated and validated for flow cytometric analysis of stimulated human peripheral blood cells. Use 5 µL (0.125 µg) per test (100 µL final volume). Cell number range: 10⁵–10⁸ cells/test.

Fluorochrome Information:
PerCP-eFluor® 710 can replace PE-Cy5, PE-Cy5.5, or PerCP-Cy5.5. It emits at 710 nm and is excited with the 488 nm blue laser. Recommended filter setup: 685 LP dichroic mirror and 710/40 band pass filter (695/40 BP acceptable alternative).

PerCP-eFluor® 710 conjugated antibodies are stable in samples fixed with freshly prepared 2% formaldehyde overnight at 4°C. Evaluate for alternative fixation protocols as needed.

Target Information

GARP (leucine rich repeat containing 32) is a type I membrane protein with 22 leucine-rich repeats and a short intracytoplasmic region. It was identified through gene expression profiling of Treg and Th cells following TCR stimulation. Since GARP is expressed in CD4⁺, CD25⁺, FoxP3⁺ cells (Tregs), it serves as a potential surface marker for Treg cells.
