Product Specific Information

Description:
Monoclonal antibody 87G recognizes human HLA-G, a nonclassical MHC class I molecule involved in immune inhibition. It recognizes both HLA-G1 and soluble HLA-G5 isoforms. HLA-G is primarily expressed in fetal trophoblast cells to protect the fetus from maternal immune attack. Expression has also been observed in certain tumors (melanoma, breast, renal carcinoma, CLL, AML, B-CLL) and in pancreatic islets, erythroid and endothelial progenitors, and adult thymic medulla. HLA-G⁺ CD4/CD8 cells in peripheral blood act as regulatory cells with distinct cytokine profiles.

Applications Reported:
For use in flow cytometric analysis.

Applications Tested:
Pre-titrated and tested by flow cytometry using stimulated monocytic cell lines. Recommended use: 5 µL (0.125 µg) per test (100 µL final volume; 10⁵–10⁸ cells/test).

Fluorochrome Information:
PerCP-eFluor® 710 can substitute for PE-Cy5, PE-Cy5.5, or PerCP-Cy5.5.

  • Excitation: 488 nm (Blue Laser)
  • Emission: 710 nm
  • Recommended filters: 685 LP dichroic mirror with 710/40 band pass (or 695/40 BP alternative).
  • Stable after fixation with 2% formaldehyde overnight at 4°C.

Filtration:
0.2 µm post-manufacturing filtered.


Target Information

HLA-G is a class I HLA heavy chain paralogue forming a heterodimer with β2-microglobulin. The heavy chain (~45 kDa) is membrane-anchored and encoded by 8 exons:

  • Exon 1: leader peptide
  • Exons 2–3: α1, α2 domains (peptide-binding)
  • Exon 4: α3 domain
  • Exon 5: transmembrane region
  • Exon 6: cytoplasmic tail

HLA-G is expressed on fetal placental cells and plays a role in immune tolerance.
HLA and MHC antibodies are critical for Immunopeptidomics, aiding in neoantigen identification via LC-MS/MS.
