Product Specific Information

The HTA125 monoclonal antibody reacts with human Toll-like receptor 4 (TLR4). The Toll receptor family consists of type I transmembrane proteins with leucine-rich extracellular domains and IL-1 receptor–like cytoplasmic domains. TLR4, together with MD-2 and CD14, forms a complex responsible for LPS recognition and signaling, playing a critical role in innate immunity and inflammation. TLR4 is primarily expressed on peripheral blood monocytes.

HTA125 has been shown to immunoprecipitate human TLR4 (~100 kDa) from transfected cells. TLR expression levels vary among donors and are generally low on monocytes and other immune cells.

Recommended Protocol:
For optimal TLR4 staining, use the lysed whole blood protocol rather than Ficoll-gradient separation, as density gradients can reduce staining intensity.

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-diluted and validated for 5 µL (0.5 µg) per test. One test = staining of 10⁵–10⁸ cells in 100 µL volume.

Fluorophore Information:
PerCP-eFluor 710 is excited by the 488 nm blue laser and emits at 710 nm. Recommended filter: 710/50 bandpass (695/40 acceptable). Protect from light to prevent photo-oxidation.

Fixation Compatibility:
Samples can be fixed in IC Fixation Buffer (#00-8222-49) or 1-step Fix/Lyse Solution (#00-5333-57) for up to 3 days at 4°C in the dark with minimal signal loss.


Target Information

TLR4 (Toll-like receptor 4) is a key receptor in pathogen recognition and activation of innate immunity. It is part of the LPS receptor complex with CD14 and LY96, mediating responses to Gram-negative bacterial lipopolysaccharide. TLR4 expression is highest in placenta and myelomonocytic cells. Mutations in TLR4 can alter LPS responsiveness. Several transcript variants exist, though their coding potential is uncertain. In TLR4-deficient mice, impaired LPS response leads to increased susceptibility to Gram-negative sepsis.
