Product Specific Information

Description:
The OKT8 monoclonal antibody reacts with the human CD8a molecule (~32–34 kDa), expressed as CD8αβ heterodimer or CD8αα homodimer. CD8a is found on thymocytes, mature T cells, and NK cells. It binds MHC class I and associates with p56lck, playing a role in T-cell development and activation.

Preliminary testing shows OKT8, RPA-T8, and HIT8a clones do not compete for binding to human peripheral blood leukocytes, indicating distinct epitopes.

Applications Reported:

• Flow cytometric analysis of human peripheral blood cells.

Applications Tested:

• Pre-titrated for flow cytometry at 5 µL (0.25 µg) per test.
• Suitable for 10⁵–10⁸ cells/test in a 100 µL final volume.

• Excitation: 405 nm
• Emission: 436 nm
• Recommended filter: 450/50 bandpass
• Ensure instrument capability for Super Bright 436 detection.

Buffer Recommendation:
When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Target Information

Cluster of differentiation 8 (CD8) is a type I transmembrane glycoprotein involved in T cell signal transduction, differentiation, and activation.
CD8 is expressed on T cells as CD8αβ heterodimer or CD8αα homodimer and functions as a co-receptor with TCR for MHC class I molecules.
It is also found on subsets of NK cells, monocytes, and dendritic cells.
Activation via MHC-I/peptide complexes triggers Lck recruitment, cytokine production, and cytotoxic T lymphocyte (CTL) activity.
Differentiation of naive CD8+ T cells into CTLs is enhanced by IL-2, IL-12, and TGF-β1.
