Product Specific Information

The CBRM1/5 monoclonal antibody recognizes an activation-specific epitope of human Mac-1 (CD11b/CD18). It binds a subset of Mac-1 molecules on neutrophils and monocytes after stimulation with chemoattractants or phorbol esters, but not on resting cells.
This epitope localizes to the I domain on the α-chain of Mac-1 near the ligand binding site. CBRM1/5 blocks Mac-1–dependent adhesion to fibrinogen and ICAM-1 and inhibits chemoattractant-stimulated adhesion of eosinophils to ICAM-1.
Low-level activation may occur during blood processing, causing minor binding to Mac-1 in unstimulated samples; however, higher expression is observed in activated cells.

Target Information

CD11b (integrin alpha-M, ITGAM) is a 165 kDa adhesion molecule that associates non-covalently with integrin beta-2 (CD18), forming the Mac-1 (CR3) complex.
CD11b is expressed on monocytes/macrophages, granulocytes, activated lymphocytes, subsets of NK and dendritic cells, and microglia. It functions as a receptor for ICAM-1, ICAM-2, ICAM-4, CD14, CD50, CD23, heparin, iC3b, fibrinogen, and Factor X, mediating cell-cell and cell-matrix interactions.
Expression levels vary by tissue and activation state, with higher expression in activated or peritoneal macrophages.
Dysfunction of CD11b is linked to autoimmune diseases such as systemic lupus erythematosus.
