Product Specific Information
The eBioCB-CALLA monoclonal antibody recognizes human CD10 (CALLA, NEP, enkephalinase, Neprilysin), a 100 kDa type II cell surface glycoprotein originally identified on acute lymphoblastic leukemia (ALL) cells. CD10 is identical to neutral endopeptidase (NEP), a Zn²⁺-dependent metallo-peptidase that regulates chemotactic and inflammatory processes through degradation of peptides such as endothelin, glucagon, gastrin, neurotensin, and bradykinin.
In B cells, CD10 regulates stromal cell-dependent B lymphopoiesis and is expressed on mature B cells in germinal centers. Other major expression sites include enterocyte brush borders, renal tubules, and glomeruli. Partial blocking between eBioCB-CALLA and MEM-78 antibodies suggests recognition of similar epitopes.
Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and tested by flow cytometry on normal human peripheral blood cells. Recommended use: 5 µL (0.125 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).
Fluorochrome Information:
Super Bright 436 is excited at 405 nm and emits at 436 nm. Recommended detection filter: 450/50 bandpass. Ensure instrument compatibility.
Staining Recommendation:
When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific interactions.
Excitation: 405 nm
Emission: 436 nm
Laser: Violet Laser
Super Bright Polymer Dyes are sold under license from Becton, Dickinson and Company.
Target Information
CD10 (Common Acute Lymphocytic Leukemia Antigen, CALLA) is a cell surface enzyme with neutral metalloendopeptidase activity that inactivates various biologically active peptides. It is expressed on lymphoblastic, Burkitt’s, and follicular germinal center lymphoma cells, immature B cells, and cells from chronic myelocytic leukemia (CML) patients. CD10 is also present on breast myoepithelial cells, bile canaliculi, fibroblasts, and highly expressed on the brush border of kidney and gut epithelial cells.
It cleaves peptides at the amino side of hydrophobic residues, inactivating peptide hormones including glucagon, enkephalins, substance P, neurotensin, oxytocin, and bradykinin. CD10 is a 100 kDa type II transmembrane glycoprotein encoded by a >45 kb gene with alternatively spliced 5’ UTRs producing four mRNA transcripts. Diseases associated with CD10 dysfunction include spinocerebellar ataxia 43 and Charcot-Marie-Tooth disease.
