• Immunohistochemistry (PFA fixed): Incubate antibody for 30 minutes at room temperature.
• For formalin-fixed tissues, heat sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 minutes at 95°C, then cool at room temperature for 20 minutes before staining.

Target Information

BCL-2 is a key regulator of apoptosis that can inhibit or promote cell death. Members of the BCL-2 family form dimers to modulate apoptosis. For example, Bag1 forms a heterodimer with BCL-2 to enhance its anti-apoptotic effect. Bax and Bak promote cytochrome c release from mitochondria, initiating apoptosis.
Constitutive expression of BCL2, such as through translocation to the Ig heavy chain locus, is associated with follicular lymphoma. In most follicular lymphomas, neoplastic germinal centers express high levels of BCL-2 alpha protein, whereas normal or hyperplastic germinal centers do not. Two transcript variants of BCL-2 differ in their C-terminal ends due to alternative splicing. Overexpression of BCL-2 is linked to cancers such as B-cell lymphoma and prostate cancer.
