Immunohistochemistry (PFA fixed): Incubate antibody for 30 minutes at room temperature.
Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.

Target Information

BCL-2 is a key regulator of apoptosis that functions to either inhibit or promote cell death.
Members of the BCL-2 family can dimerize to modulate apoptosis. For example, Bag1 forms a heterodimer with BCL-2, enhancing its anti-apoptotic effect.
Bax and Bak play critical roles in cytochrome c release from mitochondria and initiation of apoptosis. Bax exerts a pro-apoptotic effect.
Constitutive expression of BCL2, such as from translocation to the Ig heavy chain locus, is associated with follicular lymphoma.
In most follicular lymphomas, neoplastic germinal centers express high levels of BCL-2 alpha protein, whereas normal or hyperplastic germinal centers are negative.
Two transcript variants of BCL-2 differ in their C-terminal ends. Overexpression of BCL-2 has been linked to cancers such as B-cell lymphoma and prostate cancer.
