Product Specific Information
- Immunohistochemistry (PFA fixed): incubate antibody for 30 min at room temperature.
- Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 min at 95°C, followed by cooling at room temperature for 20 minutes.
Target Information
BCL-2 is a key regulator of apoptosis that functions to either inhibit or promote cell death. Members of the BCL-2 family dimerize to modulate apoptosis. For example, Bag1 forms a heterodimer with BCL-2, enhancing its anti-apoptotic effect. Bax and Bak are critical for cytochrome c release from mitochondria, initiating apoptosis. Constitutive expression of BCL-2, such as in translocation to the Ig heavy chain locus, is associated with follicular lymphoma. In most follicular lymphomas, neoplastic germinal centers express high levels of BCL-2 alpha protein, while normal or hyperplastic germinal centers are negative. Two transcript variants of BCL-2 differ in their C-terminal ends. Overexpression of BCL-2 is linked to cancers such as B-cell lymphoma and prostate cancer.
