Immunogen
8E5 was prepared by fusing Sp2/o myeloma cells with spleen cells derived from female BALB/c mice immunized with a synthetic beta-fibrin peptide sequence GHRPLDKC conjugated to MB-KLH (maleimidobenzoylated keyhole limpet hemocyanin).
Product Specific Information
Specificity:
8E5 binds to the amino-terminal region of the beta-chain of human fibrin at an epitope consisting of GHRPLDKC.
Binding occurs in both monomeric and polymeric forms of fibrin.
The epitope is accessible only after conversion of fibrinogen to fibrin via thrombin-mediated removal of fibrinopeptides A & B.
Therefore, this antibody is fibrin-specific.
Fibrin is a non-globular protein essential for blood clotting, forming an insoluble matrix with platelets at wound sites.

FIBB (Fibrinogen Beta Chain)
FIBB is the beta component of fibrinogen, a blood-borne glycoprotein composed of three pairs of nonidentical polypeptide chains.
After vascular injury, fibrinogen is cleaved by thrombin to form fibrin, the major component of blood clots.
Cleavage products of fibrinogen and fibrin regulate cell adhesion, spreading, vasoconstriction, chemotaxis, and act as mitogens.
Mutations in the FIBB gene can lead to afibrinogenemia, dysfibrinogenemia, hypodysfibrinogenemia, and thrombotic disorders.
