Target Information

The leptin (OB-R) receptor is a member of the cytokine receptor superfamily that plays a critical role in mammalian body weight homeostasis and energy balance. Ligand-induced activation of OB-R by leptin triggers downstream signaling via the JAK/STAT pathway. OB-R can dimerize extracellularly, and intracellular domain dimerization initiates signaling. Multiple isoforms exist with differing signaling capacities. The long form of OB-R is expressed in the choroid plexus and hypothalamus, regions associated with satiety and energy regulation. Mutations in OB-R (db) or leptin (ob) genes cause early-onset obesity and metabolic abnormalities such as hyperinsulinemia, insulin resistance, and altered CNS activity.
