Target Information

Matrix metalloproteinases (MMPs) are proteolytic enzymes capable of degrading connective tissue components. They share a conserved activation mechanism and metal-binding active site region and are inhibited by tissue inhibitors of metalloproteinases (TIMPs). MMPs and TIMPs play significant roles in regulating angiogenesis.

MMP2 is synthesized as a 631-amino-acid proenzyme, activated by cleavage of the first 80 amino acids, and contains propeptide, catalytic, and hemopexin domains. MMPs can degrade all components of the extracellular matrix, including collagens, fibronectin, laminin, and glycoproteins. Structurally, MMPs have a signal peptide, a propeptide, and a catalytic domain containing a conserved zinc-binding site.

Functionally, MMP2 is involved in tissue remodeling. Mutations in the MMP2 gene are associated with Winchester syndrome and Nodulosis-Arthropathy-Osteolysis (NAO) syndrome. Two transcript variants encoding different isoforms of MMP2 have been identified.
