Product Specific Information
The eBio16TFB antibody reacts with Transforming Growth Factor-beta (TGF-beta), a pleiotropic cytokine existing in five isoforms (TGF-β1–5) with 70–80% homology among them. TGF-β1 is the most abundant form, widely expressed in lymphoid organs, while other isoforms have more restricted expression. All isoforms are biologically active as disulfide-linked homodimers.
TGF-β1 is produced at high levels by platelets and also by macrophages, lymphocytes, endothelial cells, chondrocytes, and leukemic cells. Its secretion can be induced by steroids, retinoids, EGF, NGF, vitamin D3, and IL-1. TGF-β1 inhibits proliferation of various cell types including T cells and NK cells, and downregulates macrophage activity. It also blocks IL-2–mediated anti-tumor activity of lymphokine-activated killer (LAK) cells.
Recent studies show TGF-β1 plays a key role in regulatory T cell development. Dendritic cells exposed to tumors secrete TGF-β1, promoting expansion of Treg cells and inducing Foxp3 expression, leading to differentiation of CD4⁺CD25⁺ Treg cells (Ti-Treg).
Applications Reported: Suitable for use in ELISA.
Applications Tested: Used as detection antibody in sandwich ELISA for human and mouse TGF-beta, in combination with capture antibody eBioTB2F (Product #14-9943-81). Recommended concentration range: 2–6 µg/mL. Include a standard curve from 8000 pg/mL to 60 pg/mL per plate.
This antibody recognizes the mature/active form of TGF-beta1 (not associated with LAP). Most samples require acid treatment and neutralization to remove LAP prior to analysis. Some serum/plasma samples may contain low levels of free TGF-beta1 measurable without acid treatment.
Target Information
TGF-beta1, -2, and -3 are closely related proteins (70–80% sequence homology) produced by many cell types, functioning as growth and differentiation factors. The active forms are disulfide-linked homodimers.
