Product Specific Information

The C398.4A monoclonal antibody reacts with ICOS (Inducible Costimulatory molecule), also known as H4/CRP-1/AILIM, and cross-reacts with mouse and rat ICOS. ICOS is a T cell-specific activation molecule, a member of the CD28/CTLA-4 family, expressed on activated T cells. It plays a key role in T cell activation, proliferation, and humoral immune response.

ICOS binds to its ligand B7h/B7RP-1 on activated APCs, including B cells, and is involved in Th2 cell development and autoimmune regulation.
C398.4A displays strong costimulation of T cell clone proliferation, intermediate activation on activated T cells, and weak activation on resting T cells. It enhances IL-10 production from pre-activated cells expressing high levels of ICOS. The epitope recognized by C398.4A is distinct from that bound by ICOSL (does not block ICOSL binding).

Applications Reported: Suitable for flow cytometric analysis.
Applications Tested: Tested on Con A-stimulated mouse splenocytes at ≤0.125 µg/test (100 µL volume). Optimal titration recommended.

Target Information

ICOS (CD278) encodes a protein in the CD28 and CTLA-4 receptor family. It functions as a homodimer and is crucial for cell-cell signaling, immune responses, and regulation of proliferation. ICOS has been associated with disorders such as Common Variable Immunodeficiency (CVID).
