Product Specific Information

The MTS510 monoclonal antibody reacts with the mouse Toll-like receptor 4 (TLR4)/MD-2 complex. The Toll family of type I transmembrane proteins includes receptors with extracellular leucine-rich repeats and cytoplasmic domains homologous to the type I IL-1 receptor. TLR2 and TLR4 are key pattern recognition receptors involved in innate immunity and inflammation.

TLR4 forms a complex with MD-2 and CD14 to mediate LPS recognition and signaling. In mice, TLR4 is expressed by thioglycolate-elicited peritoneal macrophages, and its surface expression decreases upon LPS stimulation. Defective TLR4 genes in C3H/HeJ and C57BL/10ScCr mice result in hyporesponsiveness to LPS.

The MTS510 antibody co-immunoprecipitates MD-2 (~30 kDa) and TLR4 (~100 kDa), preferentially binding to the TLR4/MD-2 complex. In contrast, UT41 antibody binds regardless of complex formation.

Applications Reported: Flow cytometry and immunohistochemical staining of frozen tissue sections.
Applications Tested: Tested by flow cytometric analysis of mouse thioglycolate-elicited peritoneal exudate cells. Recommended ≤0.5 µg/test (100 µL final volume). Cell density may range from 10⁵–10⁸ cells/test. Optimize concentration for specific assay conditions.

Target Information

MD-2 is part of the Toll/interleukin-1 receptor (TIR) family, which includes Toll-like receptors (TLRs) that recognize pathogen-associated molecular patterns (PAMPs). TLR4, the main receptor for lipopolysaccharide (LPS), requires MD-2 binding for maximal signaling. The LPS response specificity depends on MD-2 species; human MD-2 expression in mouse cells can alter TLR4 ligand specificity.
