Product Specific Information

The eBioSN5c monoclonal antibody recognizes human CD10 (CALLA, NEP, enkephalinase, Neprilysin), a 100 kDa type II cell surface glycoprotein originally identified on most acute lymphoblastic leukemias (ALLs). CD10 acts as a Zn²⁺-dependent metallopeptidase regulating various peptides such as endothelin, glucagon, gastrin, neurotensin, and bradykinin. It plays a role in chemotactic and inflammatory processes involving neutrophils and in stromal cell-dependent B lymphopoiesis. Expression is observed on mature germinal center B cells, granulocytes, and bone marrow stromal cells.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested by flow cytometry using normal human peripheral blood cells. Use 5 µL (0.25 µg) per test (100 µL total volume; 10⁵–10⁸ cells/test).

Fluorochrome Information:
PerCP-eFluor® 710 emits at 710 nm and is excited with the 488 nm blue laser. Recommended bandpass filter: 710/50 (695/40 acceptable). Ensure instrument compatibility.

Fixation:
Samples may be stored in IC Fixation Buffer (Product # 00-8222-49) or 1-step Fix/Lyse Solution (Product # 00-5333-54) for up to 3 days at 4°C in the dark. Clone-specific performance should be verified empirically.

Target Information

CD10 (Common Acute Lymphocytic Leukemia Antigen, CALLA) is a cell surface enzyme with neutral metalloendopeptidase activity that inactivates various biologically active peptides. It is expressed on lymphoblastic, Burkitt’s, and follicular germinal center lymphomas, immature B cells in adult bone marrow, and cells from chronic myelocytic leukemia (CML) patients. CD10 is also present on breast myoepithelial cells, bile canaliculi, fibroblasts, and highly expressed on kidney and gut epithelial brush borders. It cleaves peptides at the amino side of hydrophobic residues, inactivating hormones like glucagon, enkephalins, substance P, neurotensin, oxytocin, and bradykinin. CD10 is a 100 kDa type II transmembrane glycoprotein encoded by a gene with multiple alternatively spliced 5′ UTRs without affecting the coding region.
