Description:
The OX8 monoclonal antibody reacts with rat CD8 alpha, a member of the immunoglobulin superfamily that heterodimerizes with CD8 beta to form the CD8 antigen. It is expressed on CD4+CD8+ thymocytes, MHC class I-restricted peripheral T cells, and some macrophages. CD8 binds MHC class I on antigen-presenting cells and participates in T-cell receptor signal transduction via Lck kinase.

Reported for use in flow cytometric analysis.

Applications Tested:
Tested by flow cytometric analysis of rat splenocytes. Recommended ≤0.06 µg per test (defined as staining a 100 µL cell sample). Optimal titration required for specific assay conditions.

Fluorochrome Information:
PerCP-eFluor® 710 emits at 710 nm, excited by blue laser (488 nm). Recommended filter: 710/50 bandpass (695/40 acceptable). Verify instrument compatibility.

Fixation:
Samples can be stored in IC Fixation Buffer (Product #00-8222-49) or 1-step Fix/Lyse Solution (Product #00-5333-54) for up to 3 days at 4°C in the dark with minimal signal loss.

Target Information

Cluster of differentiation 8 (CD8) is a type I transmembrane glycoprotein of the immunoglobulin receptor family. It plays critical roles in T cell signal transduction, differentiation, and activation. CD8 is expressed mainly on T cells as a heterodimer (CD8α/CD8β) acting as a co-receptor for MHC class I molecules, while CD4 acts for MHC-II. CD8α also forms homodimers on subsets of T cells, NK cells, monocytes, and dendritic cells. Upon MHC-I/peptide binding, CD8 recruits Lck kinase, leading to cytokine production, motility, and cytotoxic T lymphocyte activation. Activated CTLs are essential for pathogen and tumor clearance. Differentiation of naive CD8+ T cells into CTLs is enhanced by IL-2, IL-12, and TGF-β1.
