Product Specific Information

The OKT8 monoclonal antibody reacts with the human CD8a molecule, a 32–34 kDa cell surface receptor expressed as either a heterodimer (CD8αβ) or homodimer (CD8αα). CD8a is found on thymocytes and subsets of mature T cells and NK cells. It binds MHC class I and associates with protein tyrosine kinase p56lck, playing a role in T-cell development and activation.

Preliminary testing indicates that OKT8 and other mouse anti-human CD8 antibodies (RPA-T8 and HIT8a) do not compete for binding to human peripheral blood leukocytes, suggesting distinct epitopes.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and validated for flow cytometry of normal human peripheral blood cells. Use at 5 µL (0.25 µg) per test (100 µL final volume).

Fluorophore Information:
PerCP-eFluor 710 emits at 710 nm and is excited by a 488 nm blue laser. Recommended filter: 710/50 bandpass (695/40 acceptable). Ensure instrument compatibility.

Light Sensitivity: Protect from light to prevent photo-induced oxidation.
Fixation: Samples may be stored up to 3 days at 4°C in IC Fixation Buffer or 1-step Fix/Lyse Solution with minimal impact on brightness or FRET efficiency.

Target Information

CD8 is a type I transmembrane glycoprotein of the immunoglobulin receptor family, crucial for signal transduction and T cell differentiation. It functions as a co-receptor with TCR for MHC class I molecules. CD8α is essential for MHC-I binding and is expressed on T cells, NK cells, monocytes, and dendritic cells.

Upon MHC-I/peptide binding, CD8 recruits Lck kinase, leading to lymphokine production and cytotoxic T lymphocyte (CTL) activation. Activated CTLs are vital for pathogen and tumor clearance. Differentiation of naive CD8+ T cells into CTLs is enhanced by IL-2, IL-12, and TGF-β1.
