Target Information

Smad4 is encoded by the Smad4 gene located on chromosome 18 in humans. Also known as Smad family member 4, MAD homolog 4, or Mothers against decapentaplegic homolog 4.
Smad4 contains N-terminal MH1 and C-terminal MH2 domains involved in DNA binding and protein interactions.
Upon TGF-β or BMP ligand binding, receptor phosphorylation activates R-Smads (1, 2, 3, 5, 8), which form complexes with Smad4 and translocate to the nucleus.
Smad4 functions as a tumor suppressor by regulating genes such as Cyclin D1 (downregulated) and collagen (upregulated), affecting cell proliferation.
Dephosphorylation controls nuclear export and Smad nucleocytoplasmic dynamics.
