Target Information
TRADD (Tumor necrosis factor receptor type 1-associated DEATH domain protein)는 FAS 및 TNFR1 관련 세포자멸사(apoptotic) 신호전달의 중요한 매개체입니다. 핵 내 형태는 TRIP12에 의한 CDKN2A의 p19ARF/ARF 이소폼의 유비퀴틴화 및 분해를 억제하여 종양 억제 인자로 작용합니다. TRADD는 TRIP12와 상호작용하여 TRIP12와 CDKN2A p19ARF/ARF 이소폼 간의 결합을 방해합니다. 또한 TNFRSF1A/TNFR1의 활성화된 세포질 도메인과 결합하여 FADD와의 상호작용을 매개하는 어댑터 분자로 작용합니다. TRADD의 과발현은 TNF 유도 반응 중 세포자멸사와 NF-kappa-B 활성화를 유도합니다.
