• Recommended positive controls: HeLa, HCT116 treated with 30 µM cisplatin for 24 hr
• Predicted reactivity: Cat (84%), Sheep (82%), Rhesus Monkey (98%), Bovine (83%)
• IHC notes: Requires antigen retrieval using heat-mediated 10 mM Citrate buffer (pH 6.0) or Tris-EDTA buffer (pH 8.0)
• Store product as a concentrated solution. Centrifuge briefly prior to opening the vial.

Target Information

Protein p21 is a tumor suppressor protein that inhibits Cyclin-dependent kinases (CDK) and is transcriptionally activated by p53. It plays a key role in the cellular response to genotoxic stress as a major transcriptional target of p53.
In the nucleus, p21 binds and inhibits Cdk1 and Cdk2, blocking the transition from G1 to S phase or from G2 to mitosis after DNA damage, allowing DNA repair.
In the cytoplasm, p21 exhibits anti-apoptotic effects by binding to and inhibiting caspase 3, ASK1, and JNK.
p21 has dual roles in carcinogenesis, acting as both a tumor suppressor and an oncogene depending on context.
