Target Information

RIPK2 is a serine/threonine kinase involved in cell activation, proliferation, differentiation, and apoptosis. Upon stimulation by bacterial peptidoglycans, NOD1 and NOD2 are activated, oligomerize, and recruit RIPK2 through CARD-CARD domains. It contributes to tyrosine phosphorylation of ARHGEF2 via Src kinase, leading to NF-kappaB activation. RIPK2 undergoes autophosphorylation and Lys-63-linked polyubiquitination by E3 ligases XIAP, BIRC2, and BIRC3, facilitating MAP3K7/TAK1 recruitment and NF-kappaB activation. This signaling cascade regulates immune response, growth control, and apoptosis protection. RIPK2 also participates in T-cell receptor engagement, promoting BCL10 phosphorylation and NF-kappaB activation.
