Product Specific Information

Description:
The O323 monoclonal antibody reacts with human CD27, a lymphocyte-specific member of the TNFR superfamily. CD27 is expressed by a subset of thymocytes and virtually all mature T cells, and is upregulated upon T-cell stimulation. CD27 binds to CD70, playing an important role in T cell–B cell interaction.

Applications Reported:
The O323 antibody has been reported for use in flow cytometric analysis.

Applications Tested:
Pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells. Use 5 µL (0.25 µg) per test (final volume 100 µL). Cell number may range from 10⁵–10⁸ cells/test.

Target Information

CD27 is a 50 kDa member of the TNF receptor superfamily, including CD40 and CD30, involved in regulating cell proliferation and death. Expression of CD27 and its ligand CD70 is primarily confined to lymphocytes. CD27 is expressed as a disulfide-linked homodimer on mature thymocytes, peripheral blood T cells, and a subset of B cells. Upon T cell activation, CD27 expression increases, and soluble CD27 (sCD27) is released, serving as a prognostic marker in B cell malignancies. The CD27-binding protein SIVA is a pro-apoptotic molecule implicated in CD27-mediated apoptosis. Disorders linked to CD27 dysfunction include Lymphoproliferative Syndrome 2 and Autosomal Recessive Lymphoproliferative Syndrome.
