Product Specific Information

The 37.51 monoclonal antibody reacts with the mouse CD28 molecule, a 45 kDa homodimer expressed by thymocytes, mature T cells, and NK cells. CD28 is a ligand for CD80 (B7-1) and CD86 (B7-2) and acts as a potent costimulator of T cells.
Signaling through CD28 enhances IL-2 and IL-2 receptor expression, as well as cytotoxicity of CD3-activated T cells.

• Applications Tested: Tested by flow cytometric analysis of mouse splenocytes, ≤0.5 µg per test (100 µL final volume).
  Optimal performance should be determined empirically.
• Note: Longer incubation times (30–45 min) can enhance staining.
• Excitation: 633–647 nm; Emission: 660 nm; Laser: Red Laser.
• Filtration: 0.2 µm post-manufacturing filtered.

Target Information

CD28 antigen is a 44 kDa disulfide-linked homodimeric T cell surface glycoprotein belonging to the immunoglobulin superfamily. It is constitutively expressed on most peripheral blood T lymphocytes and serves as a critical costimulatory receptor by binding CD80 and CD86 on antigen-presenting cells.
CD28 prevents T cells from entering an anergic state or undergoing premature apoptosis. In mice, CD28 is expressed on all CD4+ and CD8+ T cells in peripheral lymphoid organs and blood. Expression is highest on immature thymocytes and decreases as cells mature.
Diseases associated with CD28 dysfunction include mycosis fungoides and Sezary’s disease.
