The 5B8 monoclonal antibody reacts with canine CD28, which is expressed by thymocytes, most T cells, and a subset of NK cells.
CD28 acts as a ligand for CD80 (B7-1) and CD86 (B7-2), serving as a potent costimulator of T cells. Signaling through CD28 enhances IL-2 and IL-2 receptor expression and increases the cytotoxicity of CD3-activated T cells.

• The 5B8 antibody has reported agonist activity.
• The 5B8 and 1C6 antibodies recognize different epitopes of canine CD28.
• Reported application: Flow cytometric analysis.
• Tested application: Flow cytometric analysis of normal canine peripheral blood cells.
• Recommended usage: 5 µL (0.25 µg) per test (100 µL final volume).

Target Information

CD28 antigen is a 44 kDa disulfide-linked homodimeric T cell-specific surface glycoprotein.
It is a member of the immunoglobulin superfamily and is constitutively expressed on most peripheral blood T lymphocytes. CD28 serves as a key costimulatory receptor, binding to CD80 and CD86 on antigen-presenting cells to deliver the second activation signal required for T cell activation.

In addition to its co-stimulatory role, CD28 prevents T cells from entering an anergic or apoptotic state.
In mice, CD28 is expressed on all CD4⁺ and CD8⁺ T cells, with expression levels varying across thymocyte developmental stages.
Dysfunction of CD28 has been associated with diseases such as mycosis fungoides and Sezary’s disease.
