Product Specific Information

The OX35 monoclonal antibody reacts with rat CD4, a co-receptor involved in T cell receptor signaling through MHC class II on antigen-presenting cells. CD4 is expressed by CD4+CD8+ double-positive and CD4+ single-positive thymocytes, as well as CD4+ helper T cells in the periphery.

Applications Tested: Flow cytometric analysis of rat splenocytes (≤ 1.0 µg/test)

Super Bright 600 is a tandem dye excited by a violet laser (405 nm) and emits at 600 nm. Recommended detection using a 610/20 bandpass filter.

Staining Buffer Recommendation:
When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Light Sensitivity: Protect vial and stained samples from light.
Fixation: Samples can be stored in IC Fixation Buffer (Product # 00-8222-49) or 1-step Fix/Lyse Solution (Product # 00-5333-57) for up to 3 days at 4°C in the dark.

Target Information

CD4 antigen is a co-receptor for MHC class II molecules and HIV. It is primarily expressed in T helper cells but can also be found in monocytes, macrophages, and dendritic cells. Tissue-level expression includes thymus, lymph nodes, tonsils, spleen, and certain non-lymphoid tissues. CD4 facilitates T-cell activation via association with Lck kinase and may mediate neuronal damage in immune-related CNS diseases. Multiple alternatively spliced transcripts have been identified (RefSeq, July 2017).
