Description:
The 85BRD monoclonal antibody reacts with human interleukin-13 (IL-13). IL-13 (NC30) is a 12.5 kDa protein secreted by activated T helper cells, CD8+ T cells, and NK cells. It suppresses macrophage cytotoxic activity, upregulates IL-1RA expression, and suppresses inflammatory cytokine expression.

Applications Reported:
This antibody is reported for use in flow cytometric analysis.

Applications Tested:
Pre-diluted and tested by flow cytometric analysis of stimulated normal human peripheral blood cells using the Intracellular Fixation & Permeabilization Buffer Set (Product # 88-8824-00).
Recommended usage: 5 µL (0.25 µg) per test (100 µL final volume).
Cell number range: 10⁵–10⁸ cells/test.

Light Sensitivity:
This tandem dye is sensitive to photo-induced oxidation. Protect vial and stained samples from light.

Fixation:
Samples can be stored in IC Fixation Buffer (Product # 00-8222-49) or 1-step Fix/Lyse Solution (Product # 00-5333-57) for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency. Clone-specific performance should be determined empirically.

Excitation/Emission:
488–561 nm excitation; 775 nm emission

Target Information

IL-13 is an immunoregulatory cytokine produced primarily by activated Th2 cells. It regulates inflammatory and immune responses by down-regulating macrophage activity, inhibiting pro-inflammatory cytokine production, and participating in allergen-induced asthma independent of IgE and eosinophils.
It influences B cell maturation, upregulates CD23 and MHC class II expression, and promotes IgE isotype switching. Structurally related to IL-3, IL-4, IL-5, and GM-CSF, the IL-13 gene is located near IL4 within the cytokine cluster on chromosome 5q.
