NAD-dependent protein deacetylase sirtuin-1 (SIRT1) links transcriptional regulation directly to intracellular energetics and coordinates multiple cellular functions such as cell cycle, DNA damage response, metabolism, apoptosis, and autophagy.
SIRT1 modulates chromatin function through histone deacetylation and promotes changes in histone and DNA methylation, leading to transcriptional repression. It is essential for skeletal muscle cell differentiation and mediates nutrient-dependent inhibition of myoblast differentiation involving AMPK and NAMPT pathways.
