The monoclonal antibody TB28-2 recognizes the kappa light chain of human immunoglobulin. The Ig kappa chain (~22 kDa) consists of one variable and one constant region. Two kappa chains together with heavy chains form the complete immunoglobulin structure. Expression of Ig kappa is typically found on more than half of B lymphocytes.
Applications Reported: Suitable for flow cytometric analysis.
Applications Tested: Tested by flow cytometric analysis of normal peripheral blood cells. Recommended usage ≤ 0.25 µg/test (100 µL total volume). Cell number should be optimized empirically (10⁵–10⁸ cells/test). Titrate for best assay performance.
Target Information
Immunoglobulins consist of two heavy and two light chains (kappa or lambda, ~22.5 kDa). Kappa and lambda chains differ by antigenic properties of their constant regions. The kappa:lambda ratio is approximately 70:30. Antibodies to kappa light chain are useful for identifying leukemias, plasmacytomas, and certain non-Hodgkin’s lymphomas. Demonstration of clonality in lymphoid infiltrates indicates malignancy.
