Target Information
p21 (CDKN1A) is a tumor suppressor protein that inhibits cyclin-dependent kinases (CDK) and is transcriptionally activated by p53. It plays a key role in the cellular response to genotoxic stress and acts as a major transcriptional target of p53.
In the nucleus, p21 binds and inhibits CDK1 and CDK2, blocking the transition from G1 to S phase or G2 to mitosis after DNA damage, allowing DNA repair.
In the cytoplasm, p21 exhibits anti-apoptotic effects by binding and inhibiting caspase 3 and apoptotic kinases ASK1 and JNK.
p21 shows dual roles in carcinogenesis, functioning as both a tumor suppressor and an oncogene depending on the cellular context.
