Target Information
Protein p21 is a tumor suppressor protein that inhibits Cyclin-dependent kinases (CDKs) and is transcriptionally activated by p53.
It plays a key role in the cellular response to genotoxic stress and acts as a major transcriptional target of p53.
In the nucleus, p21 binds and inhibits CDK1 and CDK2, blocking cell cycle progression at G1/S or G2/M after DNA damage to allow DNA repair.
In the cytoplasm, p21 has anti-apoptotic functions by binding and inhibiting caspase 3, ASK1, and JNK.
Thus, p21 exhibits dual roles in carcinogenesis, functioning both as a tumor suppressor and as an oncogene depending on context.
