Product Specific Information

The MIH5 monoclonal antibody reacts with mouse B7-H1 (PD-L1), a member of the B7 family (~40 kDa) and Ig superfamily. B7-H1 is expressed on leukocytes including T, B, NK, and DC cells, and acts as a ligand for PD-1. Interaction of PD-1 with PD-L1 or PD-L2 inhibits T and B cell responses. MIH5 is a blocking antibody.

Applications Reported:
Used for flow cytometric analysis, immunohistochemical staining (frozen sections), and blocking in functional assays.
For functional assays, use Functional Grade Purified MIH5 (Product #16-5982).
Fluorochrome-conjugated MIH5 is recommended for flow cytometry.

Applications Tested:
Tested by flow cytometric analysis of mouse splenocytes. Optimal use ≤0.25 µg/test (100 µL volume). Cell number: 10⁵–10⁸ cells/test. Titrate for optimal performance.

Target Information

Programmed death receptor ligand 1 (PD-L1, B7-H1) binds to PD-1, negatively regulating TCR signaling, reducing proliferation and cytokine production. PD-L1 is expressed on many murine and human cancers and upregulated by IFN-γ, suggesting a role in tumor immune evasion.
