Product Specific Information

The monoclonal antibody 439-9B recognizes human CD104 (Integrin beta 4), a 202 kDa subunit forming a laminin receptor with integrin alpha 6 (CD49f). CD104 is expressed on epithelial and tumor cells, involved in cell-matrix and cell-cell interactions, and plays a role in tumorigenicity. This antibody cross-reacts with bovine and does not affect adhesion of human colon adenocarcinoma cells.

Applications Reported:
Suitable for flow cytometry, immunoprecipitation, immunoblotting (nonreducing conditions), and immunohistology of frozen and paraffin-embedded sections.

Applications Tested:
Validated in flow cytometric analysis of A549 cells at ≤1 µg/test (100 µL volume). Cell count range: 10⁵–10⁸ cells/test. Antibody titration is recommended for optimal assay performance.

Target Information

Integrins are heterodimeric transmembrane glycoprotein receptors composed of alpha and beta subunits. They mediate cell-matrix and cell-cell adhesion and regulate gene expression and cell growth. The integrin beta 4 subunit forms a receptor for laminins, often associating with alpha 6 subunit, and plays a critical role in invasive carcinoma biology. Mutations in this gene are linked to epidermolysis bullosa with pyloric atresia. Multiple splice variants exist.
