Product Specific Information

Description:
The 2E7 monoclonal antibody reacts with mouse CD103 (Integrin alpha E). CD103 associates with integrin beta7 and is expressed on intraepithelial lymphocytes, a subset of peripheral lymphocytes, and dendritic epidermal T cells (DEC). E-cadherin binds to CD103, mediating lymphocyte homing to intestinal epithelium.

Applications Reported:
Used for flow cytometry, immunoprecipitation, and immunohistochemical staining of frozen tissue sections. Also applicable in in vitro functional studies. For functional assays, use Functional Grade purified 2E7 (Product #16-1031).

Applications Tested:
Tested by flow cytometric analysis of mouse splenocytes. Recommended ≤1 µg/test (100 µL final volume). Cell count may range from 10⁵–10⁸ cells/test. Antibody titration recommended for optimal performance.

Target Information

CD103 (ITGAE, Integrin alpha E) is expressed on mucosa-associated T lymphocytes and activated TGF β-1 cells. It includes a 150 kDa alpha chain and a 120 kDa beta chain. CD103 is expressed by 0.5–2% of resting lymphocytes in peripheral blood and lymphoid organs. It stains a few bone marrow cells and rarely peripheral blood lymphocytes.

CD103 belongs to the integrin family, composed of alpha and beta chains. ITGAE encodes an I-domain-containing alpha integrin, which undergoes post-translational cleavage yielding disulfide-linked heavy and light chains. Together with beta7 integrin, CD103 forms the E-cadherin binding integrin known as human mucosal lymphocyte-1 antigen. It is preferentially expressed in intestinal intraepithelial lymphocytes (IEL) and may function in adhesion and activation.
