• Immunohistochemistry (PFA fixed): incubate antibody for 30 min at room temperature.
• Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 min at 95°C, followed by cooling at room temperature for 20 min.

Target Information

Granzyme B is a member of the granzyme serine protease family, found in the granules of cytotoxic T cells and NK cells. It is also known as CGL1 (cathepsin G-like-1) and CTLA-1 (cytotoxic T lymphocyte-associated serine esterase 1). Granzyme B is crucial for inducing apoptosis in target cells through caspase activation and mitochondrial pathways. It cleaves caspase-3 and Bid protein, initiating DNA fragmentation. Granzymes are stored in specialized lytic granules of cytotoxic T lymphocytes (CTLs) and NK cells. Several granzymes (A–G) have been identified in mice, while fewer have been characterized in humans.
