For reconstitution, add 100 µL distilled water to reach a final antibody concentration of approximately 1 mg/mL.
For conjugation experiments, perform an additional desalting step using Zeba Spin Desalting Columns (7K MWCO, 0.5 mL, Product #89882).

Target Information

CD163 (M130 antigen, Ber-Mac3, Ki-M8, SM4) is a 130 kDa membrane glycoprotein belonging to the scavenger receptor cysteine-rich superfamily. It acts as a receptor for the hemoglobin-haptoglobin complex, protecting tissues from oxidative damage and contributing to iron recycling via endocytosis and heme breakdown.

CD163 is expressed on human monocytes and macrophages, mainly in the late phase of inflammation. It is present on all circulating monocytes and most tissue macrophages except those in mantle zones and germinal centers. Expression is higher on CD16⁺ monocytes and can be upregulated by IL-10, IL-6, and dexamethasone. LPS and PMA induce shedding of CD163 into plasma or supernatant.

CD163 binds hemoglobin/haptoglobin complexes in a calcium- and pH-dependent manner, showing higher affinity for multimeric haptoglobin (HP1F phenotype) compared to dimeric haptoglobin (HP1S phenotype). It also triggers intracellular signaling cascades involving tyrosine kinase-dependent calcium mobilization, inositol triphosphate production, and secretion of IL-6 and CSF1.
