Target Information

CD163 (M130 antigen, Ber-Mac3, Ki-M8, SM4) is a 130 kDa membrane glycoprotein belonging to the scavenger receptor cysteine-rich superfamily. It serves as a receptor for the hemoglobin-haptoglobin complex, protecting tissues from oxidative damage and facilitating iron recycling through endocytosis and heme breakdown.

CD163 is expressed exclusively on human monocytes and macrophages, predominantly during the late phase of inflammation. It is found on all circulating monocytes and most tissue macrophages, except those in mantle zones and germinal centers of lymphoid follicles, interdigitating reticulum cells, and Langerhans cells. Expression is high on CD14+, CD64+, and CD16+ monocytes, and is upregulated by IL-10, IL-6, and dexamethasone. CD163 shedding is induced by LPS and PMA.

The receptor binds hemoglobin/haptoglobin complexes in a calcium- and pH-dependent manner, showing higher affinity for multimeric haptoglobin (HP1F) complexes. CD163 signaling triggers tyrosine kinase-dependent calcium mobilization, inositol triphosphate production, and secretion of IL6 and CSF1.
