Product Specific Information

Description:
Monoclonal antibody HP-F1 recognizes CD85j (ILT2, LILRB1, LIR1), a member of the ILT/LIR/MIR family. CD85j is a single transmembrane glycoprotein with four ITIMs that signal via SHP-1. It is expressed on myeloid cells (monocytes, dendritic cells) and some lymphoid subsets (NK, T, B cells). Expression correlates with leukemias (ALL, CLL) and viral infections such as HIV, EBV, and CMV. Ligands include MHC Class I molecules (HLA-G, A, F, B27, E, F). HP-F1 reduces CD16-dependent cytolytic activity of NK cells.

Each product includes:

  • 1 vial of NovaFluor conjugate
  • 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-diluted and validated for flow cytometry of normal human peripheral blood cells. Recommended use: 5 µL (0.25 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Note:
NovaFluor dyes are incompatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for optimal performance.

CellBlox Plus Blocking Buffer (Cat. No. C001T06F01):
Required for optimal staining with NovaFluor conjugates. Add 5 µL per sample to antibody cocktails before combining with cells. For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

NovaFluor Conjugates:
Based on Phiton™ technology with engineered nucleic acid dye structures, designed to minimize spillover and spread.

Target Information

This gene belongs to the leukocyte immunoglobulin-like receptor (LIR) family located at 19q13.4. The encoded protein (subfamily B) includes extracellular immunoglobulin domains, a transmembrane region, and ITIM motifs. It binds MHC class I molecules on antigen-presenting cells, transducing inhibitory signals to regulate immune response, inflammation, and cytotoxicity. Multiple transcript variants exist.
