Preparation:
Produced from sera of goats immunized with highly pure Recombinant Human IL-17A. Anti-Human IL-17A-specific antibody was purified by affinity chromatography employing an immobilized Human IL-17 matrix.

Sandwich ELISA:
To detect Human IL-17A by sandwich ELISA (using 100 µL/well antibody solution), a concentration of 0.5–2.0 µg/mL of this antibody is required.
This antigen affinity purified antibody, in conjunction with PeproTech Biotinylated Anti-Human IL-17A (500-P07Bt) as a detection antibody, allows the detection of at least 0.2–0.4 ng/well of Recombinant Human IL-17A.

Western Blot:
To detect Human IL-17A by Western Blot analysis, this antibody can be used at a concentration of 0.1–0.2 µg/mL.
Used in conjunction with compatible secondary reagents, the detection limit for Recombinant Human IL-17A is 1.5–3.0 ng/lane, under either reducing or non-reducing conditions.

Target Information

Interleukin-17A (IL-17A, CTLA-8) is a CD4+ T cell-derived cytokine that promotes inflammatory responses in cell lines and is elevated in rheumatoid arthritis, asthma, multiple sclerosis, psoriasis, and transplant rejection.
IL-17A is a 32 kDa, disulfide-linked homodimer consisting of 136 amino acids, and a member of the six-species family of proteins (IL-17A–17F). It signals through the IL-17 receptor (IL-17R/CDw217).
High levels of IL-17A homodimer are produced by activated peripheral blood CD4+ T-cells. IL-17A enhances ICAM-1 expression in fibroblasts and stimulates epithelial, endothelial, or fibroblastic cells to secrete IL-6, IL-8, G-CSF, and PGE2.
In the presence of IL-17A, fibroblasts can sustain the proliferation of CD34+ hematopoietic progenitors and induce maturation into neutrophils. Mouse, rat, and human IL-17A can induce IL-6 secretion in mouse stromal cells, indicating all homologs recognize the mouse receptor.
IL-17A regulates NF-κB and MAP kinase activities, stimulates IL-6 and COX-2 expression, and enhances nitric oxide (NO) production.
